Ezio and the renassaince will return, as master of the order ready to take on Rome. And the best news it's coming out Fall this year. Personally I'm liking the possibilities that can come out of this, so until I hear more, I like the look of this.

The whole customizable character seems interesting, but how much emphasis will be placed upon the multiplayer? I hope that they release the game with equal emphasis, like you can go and do some solo work just as Ezio, and then if you want to you can go and do some missions which require more people. And sometimes theres mission arcs which require both some solo work and some teamwork. Just as long as they keep the Animus out of it. What do you people think of the expansion of Rome as a city? I think that Venice was amazing in AC2 and if they can get the same sense of culture and livelyness that would be great.
I think Desmond and the Animus will be kept in, Because his voice actor said he is working with a project. Brotherhood could be this project he was talking about.
I dunno why but I'm thinking it might be like MGO but with Assassin's Creed gameplay instead of MGS gameplay. That'd be pretty cool to be honest but I wouldn't mind having a large multiplayer partially free roaming (like unlocking places or changing to different segments of the map like in Bloodlines) mission based game set in Rome.
Could you please explain what MGO and MGS is for the people who don't understand (me) please? Well if they put the animus in that will suck. I see the animus just as a way for them to have a health bar... seriously the game series could all be acomplished just as well without Desmond being a part of the equation... by having the animus existing they have overcompletecated things. Also if they intend to have the animus a factor again, will the whole multiplayer thing have a animus element to it? The reason why I ask is that allthough not every character in the series has a decendant whos history is monitored by an animus the creators probably have to give a reason why you can suddenly change characters...
MGS is Metal Gear Solid, an espionage series and MGO is an online version of it and incoporated all the same gameplay elements. It was very well developed and well received. If Brotherhood is similar to that i think it will be quite good. I think that the Animus could be quite integral in the story. If you look back to AC2 there were multiple Animus machines around Abstergo, and I'm guessing that there may be other participants in their experiments, volunteers or otherwise. Just because Desmond escaped doesn't mean that Abstergo stopped testing others.
Out of all the spinoff games, not one has featured either Desmond or the animus. Well the information we have on Brotherhood states: So you play as both Ezio and your custom made character right? The animus can not be used in this game because you play as different characters. The animus is the reason why we can go from Desmond to Altair or Ezio. The sudden transition from Desmond to some victim of Abstergo would hardly any reasoning behind it compared to the reasoning behind the link between you, Desmond and his ancestors.
Desmond and the Animus will still be there when you play as Ezio even if you don't actually see them since that's where the HP bar and the map and all that other stuff come from. But when playing as a custom character you won't be Desmond but someone else. You'll never see the actual person or the Animus either but because of all the HUDs you'll know that someone is viewing the past with the Animus. Basically, it's yourself in the Animus when your a custom character unless specified otherwise.
