Well, Nia is a pretty plot-important character, and since you've never seen Gurren Lagann, I don't advise playing her. As for Kiyoh, I guess you could play her. You may want to ask DT, though, just to be safe.

Of those four I'd choose Kiyoh, but you could always choose someone from a completely different anime?
Besides, as DT can tell you, flat-chested isn't a bad thing. :3
